Is M350 RTK worth upgrading from M300?

For new purchases, yes. IP55 weather protection (vs IP45) and O3 Enterprise transmission to 20 km FCC (vs 15 km) justify the upgrade. If your M300 fleet is working well, there is no urgent need to replace it.

Are M300 payloads compatible with M350?

Most, but not all. DJI lists the Zenmuse L2, P1, H30 and H30T for both aircraft. The Zenmuse L3 is the exception — DJI's L3 spec page lists the Matrice 400 as its only supported aircraft. The Zenmuse L1 is listed for the Matrice 300 RTK only. On the H20 series DJI contradicts itself: the H20T spec table lists the Matrice 300 RTK, while DJI also lists the series under the Matrice 350 RTK.

Which has better flight time?

Both publish 55 minutes with no payload. DJI's M300 manual also publishes measured figures with a 700 g load: 45 minutes forward flight and 43 minutes hovering. Plan real missions against payload-loaded numbers, not the no-payload headline.
